and today I would like to discuss
antimicrobial preservatives for
multi-dose systems
selecting a preservative system for you
for your formulation can be a very
complex process
you would have to consider other
ingredients in your formulation and the
interaction with your preservative
pH is of course a very important aspect
and can have an influence on your
preservative and how it acts against
microorganisms
you would have to think generally about
the stability of your formulation
very importantly you want your
preservative to reduce microbial
populations and avoid regrowth
subsequently
you wanted to preserve your formulation
and also
avoid contamination with patients
pharmacopeia such as the European and
the U.S from akupe have established
standards to verify the efficacy of
preservative systems
so these pharmacopoeel guidances would
require you to test your product your
preservative for efficacy against a wide
range of macroorganisms that includes
gram positive and grab negative bacteria
fungi and yeasts however not viruses
this is not a requirement for
preservative efficacy testing
you would be including microorganisms
which for example as a spiritualist
Niger
staphylococcus pseudomonas so these are
standards in the pharmaceutical
guidances
however you can also add additional
microorganisms if you find it relevant
it can be broken Dahlia for example that
you would like to test for
you can find many different preservative
systems available on the market today
that are used very standard in
Pharmaceutical formulations
we're talking about alcohols acids
quarter near ammonium compounds and
parabens are only a few examples
we partnered with the cro in the UK that
are specialized in formulating nasal
sprays and nasal Solutions and we asked
them to compare benzoconium chloride to
other standard preservatives on the
market
what we observed from this study is that
very low concentrations of benzoconium
chloride are required to obtain the
desired efficacy compared to the other
preservatives
another aspect that is interesting here
as well is that some preservatives such
as parabens benzoic acid and benzyl
alcohol are volatile
which benzoconium chloride is not
benzirconium chloride is non-volatile
it's extremely stable
it is effective at a very wide PH range
in fact the widest PH range compared to
the other preservatives
and it is a very wide spectrum effective
preservative
which makes it an ideal preservative
system for your formulations
